General Description

GranuCult^®Tryptose sulfite cycloserine agar (base) is intended for the isolation and differentiation ofClostridium perfringens. The superior nutrient base provides optimal conditions for the development of Clostridia. Colonies producing hydrogen sulfide are characterized by blackening due to the reaction with sulfite and iron salt. Cycloserine inhibits the accompanying bacterial flora and may cause the colonies that develop to remain smaller. It also reduces diffusion and thus the disturbing blackening around theClostridium perfringenscolonies. TSC agar with MUP contains a fluorogenic substrate for the alkaline and acid phosphatase in addition to cycloserine 4-Methylumbelliferylphosphate (MUP). The acid phosphatase is a high specific indicator forClostridium perfringens. It splits the fluorogenic substrate MUP, forming 4-methylumbelliferone, which can be identified by its fluorescence in long wave UV light. Thus, a strong suggestion for the presence ofClostridium perfringenscan be obtained.

Application

GranuCult^®Tryptose sulfite cycloserine agar (base) is intended for the isolation and differentiation ofClostridium perfringensfrom food and animal feed, water and other materials. TSC agar with MUP can be prepared usingClostridium perfringensSupplement (1.00888) to detect acid phosphatase by its fluorescence under UV light.
