Takara NucleoMag Pathogen \4 x 96 Preps


The NucleoMag Pathogen kit is designed for the isolation of viral RNA and DNA and bacterial DNA from cell-free body fluids such as serum or plasma, blood, homogenized tissue sample suspensions, stool sample suspensions, and swab washes. This kit provides reagents and magnetic beads for isolation of 4 x 96 samples.

Sensitivity screening for pathogen detection in human feces samples. Dilution series of 10-3–10-4of feces samples were prepared for sensitivity screening using three replicates. Nucleic acid extraction was performed with the NucleoMag Pathogen kit and the competitor kit "R". PCR analysis was performed using the RIDA GENE Viral Stool Panel I (R-Biopharm) and RealStar Clostridium difficile PCR Kit 1.0 (Altona Diagnostics).
Viral RNA from 19 confirmed SARS-CoV-2-positive swab samples was extracted in duplicate using NucleoMag Pathogen and a competitor kit (Q). Viral RNA was quantified using a SARS-CoV-2-specific qRT-PCR assay (qScript™ XLT One-Step RT-qPCR ToughMix® + nCoV IP4 assay; Institute Pasteur, Paris). For each sample tested, the resulting CT values indicated that the NucleoMag Pathogen kit performed as well as (3/19 samples), or better than the competitor kit (16/19 samples). For two samples tested, detection of SARS-CoV-2 RNA was possible with NucleoMag Pathogen but not with the competitor kit.
Dilution series were prepared for three different sample types (nasal swab, oral swab, saliva) using inactivated SARS-CoV-2 virus. Viral RNA was extracted using NucleoMag Pathogen on a KingFisher Flex system and the RNA was quantified by qRT-PCR (AgPath-ID™ One-Step RT-PCR mix + nCoV IP4 assay, Institute Pasteur, Paris). For each sample type, viral RNA was detected consistently and reliably over a 5-log range of dilutions.
Viral RNA was extracted from 200-μl inputs of diluted SARS-CoV-2-positive saliva samples in duplicate using NucleoMag Pathogen (MN) and the MagMAX™ Viral RNA Isolation Kit (T), respectively, and quantified by qRT-PCR. For each dilution analyzed, NucleoMag Pathogen provided superior assay sensitivity as compared to the MagMAX kit.
