DLin-MC3-DMA is an ionizable cationic amino lipid (apparent pKa = 6.44) that has been used in combination with other lipids in the formation of lipid nanoparticles (LNPs).1 Administration of Factor VII siRNA in DLin-MC3-DMA-containing LNPs reduces plasma Factor VII protein levels in mice. Formulations containing DLin-MC3-DMA have been used in the development of LNPs for siRNA delivery in patients with hereditary transthyretin-mediated amyloidosis.
