Cayman 10074-A4

10074-A4 is an inhibitor of the protein-protein interaction between c-Myc and the c-Myc associated protein Max.¹ It inhibits c-Myc and Max heterodimerization when used at a concentration of 5 µM and reduces the proliferation of Rat1 and TGR-1 rat fibroblasts but not TGR-1 Myc knockout fibroblasts at 32 µM. 10074-A4 (32 µM) reduces the expression of the gene encoding ornithine decarboxylase (ODC), a c-Myc and Max target gene, and induces cell cycle arrest at the g₀/g₁ phase in Myc-overexpressing Rat1 cells. Preincubation of 10074-A4 (32 µM) with Myc-overexpressing Rat1 cells in vitro reduces tumor growth of the same cells in a mouse xenograft model.
