Interleukin 22 is a type 2 cytokine (member of the IL10 family) and plays an essential role in innate protective immunity against extracellular pathogens. It induces pro-inflammatory responses, allows production of antimicrobial peptides. IL-22 is also involved in wound-healing and regeneration of epithelial cells. Human IL-22 is produced on inflammation site by Th17 CD4+ cells, activated T-cells, Th1 and NK cells. After being secreted, it targets non-hematopoietic cells and binds to IL22R that is restricted to tissue-resident cells (fibroblasts, epithelial cells). Downstream signaling involves phosphorylation of Jak1 and Tyk2 and subsequent activation of STAT3.
