Revvity Interleukin-17 (IL-17), a pro-inflammatory cytokine produced in large amounts exclusively by T cells, is part of a family of at least six members (IL-17 A to F) of the IL-17 family. IL-17A is a glycosylated homodimeric protein composed of two 133 amino acid A chains bound by two disulfide bonds. It acts on fibroblasts and macrophages by binding to its receptors to induce the secretion of other pro-inflammatory mediators such as TNF-?, IL-1?, IL-6, IL-8. IL-17 plays critical roles in autoimmune conditions such as lupus erythematosus, rheumatoid arthritis, psoriasis, inflammatory bowel disease, multiple sclerosis, and inflammatory skin diseases. As such, inhibitors have been developed as possible treatments for many of these diseases. IL-17 is also a target for anti-inflammatory therapies to improve recovery post-stroke and to reduce the development of skin cancer. Revvity AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity.
