General description
293T cell line is a stable clone derivative of the human embryonic kidney (HEK) 293 cell line.[1] It expresses the large T antigen of simian virus 40 (SV40).[2]
Cell Line Description
The 293T cell line was created in the laboratory of Michele Calos by transfection of a sub-line of adenovirus-immortalized human embryonic kidney cells with a gene encoding the SV40T-antigen and a neomycin resistance gene. The cell line is competent for replication of vectors carrying the SV40 origin of replication. The line also has favourable tissue culture, transfection, DNA replication, gene expression, and protein production properties. It gives high titres when used to produce many viral vectors such as oncoretroviruses and lentiviruses.
Application
293T cell line human has been used:
- for lentiviral particle production[3]
- in the expression of eukaryotic proteins[4][5]
- for small interfering RNA (siRNA) transfection[6]
- for human immunodeficiency virus (HIV) transfection for pseudoviruses production[7]
- as a host for the transfection of pLKO.1.-TRC-ACE2 plasmid to study the localization and role of NL63-N, a nucleocapsid protein of human coronavirus NL63, in cell cycle progression[8]
Biochem/physiol Actions
Due to the presence of T antigen, 293T cell line favors simian virus 40 (SV40) plasmid replication as well as high protein expression.[2] It also possesses stem cell properties.[2] HEK293T is used for large-scale vector production, displays increased cell growth and is efficient for transfection experiments when compared to HEK293 cells.[9]
