Lactadherin (UniProt: Q08431), also known as Breast Epithelial Antigen BA46, HMFG, MFGM, Milk Fat Globule-EGF Factor 8 (MFG-E8), SED1, Lactadherin Short Form, and Medin, is encoded by the MFGE8 gene (Gene ID: 4240) in humans. MFG-E8 is a 46 kDa secreted glycoprotein that is widely expressed across various cell types. At its N-terminal region, MFG-E8 contains two EGF-like repeats (amino acids 2–41 and 44–88). The second EGF repeat includes an RGD integrin-binding domain, which facilitates its interaction with and activation of Vβ3 and Vβ5 integrins. Additionally, MFG-E8 possesses two C-terminal F5/8 type C domains, with the second domain exhibiting high affinity for binding to phosphatidylserine (PS) exposed on apoptotic cells, exosomes, and enveloped viruses. This structural composition enables MFG-E8 to function as a bridging molecule in multiple biological processes. It mediates the clearance of PS-exposing apoptotic cells by integrin-expressing phagocytes (macrophages and dendritic cells) during efferocytosis. It also plays a role in mammary gland involution at the end of lactation by facilitating the interaction between Milk Fat Globules (MFGs) and epithelial cells. Notably, patients with systemic lupus erythematosus (SLE) often exhibit elevated levels of lactadherin in the serum. Moreover, the major component of aortic medial amyloid that occurs in individuals over the age of 60 is a 5.5 kDa C-terminal fragment of lactadherin, known as Medin. (Ref: Hanayama, R., et al. ELISA Analysis: A representative lot detected MFG-E8 in ELISA application (Yamaguchi, H., et al. (2008). J Leukoc Biol. 83(5):1300-7).
