General description
Primary Sjögren′s syndrome (PSS), a chronic autoimmune disorder, causes a variety of problems in patients such as dry mouth, dry eyes, and other problems relating to lacrimal and salivary gland disfunction which are often known as sicca symptoms. The prevalence of PSS is hard to understand due to continual changes in disease criteria. It is still considered a highly relevant disease to understand and is considered the second most common autoimmune disease. PSS typically affects females in their 40s or 50s. Criteria for classification of PSS are based on a sum of factors such as unstimulated whole saliva flow rate and the presence of anti-Ro/SSA antibodies. Neurological conditions are also common features of PSS although their manifestations vary. Although PSS is commonly associated with its sicca related symptoms, some patients do not present sicca symptoms and may never develop them in their disease progression and manifestation.The potential therapies for salivary gland disfunction remain rather limited. This is likely due to the lack of cell models that appropriately represent the related diseases such as PSS and sicca.1 Researchers often rely on cancer cell lines or short-term cultures of primary salivary gland epithelial cells (SGECs). The iSGEC (immortalized Salivary Gland Epithelial Cell) group of cell lines appear to be the first reliable immortalized human primary SGEC lines derived from multiple female patient′s labial salivary gland biopsies. These cell lines were developed to mimic conditions of salivary gland disfunction. The pSS1 cell line was developed from a salivary gland biopsy of a patient who was affected by Primary-Sjögren′s Syndrome (PSS). The cell lines were characterized for pro-acinar and epithelial cell markers and are expected to secrete α-amylase upon β-adrenergic stimulation. Application
- Each vial contains > 1X106 viable cells.
- Cells are tested negative for infectious diseases by a Essential CLEAR Panel by Charles River Animal Diagnostic Services.
- Cells are verified to be of human origin and negative for interspecies contamination from mouse, rat, Chinese hamster, Golden Syrian hamster, and nonhuman primate (NHP) as assessed by a Contamination Clear panel by Charles River Animal Diagnostic Services
- Cells are negative for mycoplasma contamination.

Preparation Note
iSGEC-pSS1 cells should be stored in liquid nitrogen. The cells can be cultured for at least 10 passages without significantly affecting cell marker expression and function.
