Claudin-3 (UniProt: O15551; also known as Clostridium perfringens enterotoxin receptor 2 (CPE-R 2; CPE-receptor 2), Rat ventral prostate.1 protein homolog (hRVP1)) is encoded by the CLDN3 (also known as C7orf1, CPETR2) gene (Gene ID: 1365) in human. Claudin-3 (CLDN3) is a barrier forming member of the claudin family of integral membrane proteins that are essential components of tight junctions in epithelial and endothelial cells. Claudins organize into tight junction strands that form a meshwork of complex multi-protein assembly. CLDN3 and CLDN1 play a role in paracellular diffusion barriers against small and large solutes. Size and charge selective ion/water channels are formed within strands from other members of the claudin family, such as CLDN2 CLDN10, CLDN15 and CLDN16. Alterations in CLDN3 expression have been implicated in several diseases, including cancers, where it is often upregulated and associated with tumor progression and metastasis. Quality Control Testing

Evaluated by Western Blotting in T-47D cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CLDN3 in T-47D cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CLDN3 in lysates from A549 cells, HT-29 cells, and human colon tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CLDN3 in human colon tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with CLDN3 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Clone 2K5 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects CLDN3. It targets an epitope within 14 amino acids from the C-terminal half.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
