Protein-lysine 6-oxidase (UniProt: P28300; also known as EC:1.4.3.13, Lysyl oxidase) is encoded by the LOX gene (Gene ID: 4015) in human. Lysyl oxidase (LOX) is an essential enzyme involved in the post-translational modification of collagen and elastin, crucial for the structural integrity and function of connective tissues. LOX is synthesized as a proenzyme and subsequently processed into its active form, which contains a copper-binding site and a lysine tyrosylquinone cofactor essential for its enzymatic activity. LOX catalyzes the oxidative deamination of lysine residues in collagen and elastin, facilitating the formation of covalent cross-links that stabilize the extracellular matrix. LOX is expressed in various tissues, with particularly high levels in the skin, lungs, and cardiovascular system. LOX is secreted into the extracellular space. Dysregulation of LOX activity is implicated in several diseases; overexpression is associated with fibrotic disorders due to excessive cross-linking and stiffening of tissues, while underexpression can lead to weakened connective tissues, as seen in conditions like Ehlers-Danlos syndrome. Additionally, LOX is involved in cancer progression, where it can promote tumor invasion and metastasis by remodeling the extracellular matrix. Quality Control Testing

Evaluated by Western Blotting in rat colon tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected LOX in rat colon tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected LOX in lysate from mg-63 conditioned media and from rat colon tissue.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LOX in Jurkat cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LOX in human kidney and human pancreas tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with LOX peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.

Clone 1H19 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects LOX. It targets an epitope within 19 amino acids from the C-terminal half.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage.
