General description
Early growth response protein 3 (UniProt: Q06889; also known as EGR-3, Zinc finger protein pilot, PILOT) is encoded by the EGR3 (also known as PILOT) gene (Gene ID: 1960) in human. EGR-3 is a transcription factor belonging to the EGR family, characterized by its three zinc finger motifs that enable DNA binding and regulation of gene expression. The zinc fingers facilitate the recognition of specific DNA sequences, allowing EGR-3 to modulate the transcription of target genes involved in cellular proliferation, differentiation, and response to external stimuli. EGR-3 is expressed in various tissues, including the brain, immune cells, and muscle tissues, where it plays a critical role in neuronal development, immune response, and muscle regeneration. Dysregulation of EGR-3 expression has been implicated in several diseases. In the nervous system, altered EGR-3 activity is associated with neurodegenerative diseases and psychiatric disorders, such as schizophrenia, due to its role in synaptic plasticity and neuronal survival. In the immune system, EGR-3 is involved in T-cell activation and proliferation, with aberrations potentially contributing to autoimmune diseases. Evaluated by Western Blotting in U-937 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected EGR-3 in U-937 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected EGR-3 in mouse and rat brain tissue lysate, and a 1:1,000 dilution in MCF7 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with EGR-3 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected EGR-3 in human cerebral cortex and human small intestine tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected EGR-3 in MCF7 cells.

Biochem/physiol Actions
Clone 1N11 is a ZooMAb® rabbit recombinant monoclonal antibody that detects EGR-3. It targets an epitope within 25 amino acids from the internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
