General description
Soft tissue sarcomas (STS) are a rare type of tumor made up of mixed cell populations with mesenchymal features. They are classified into many histological subtypes such as undifferentiated pleomorphic sarcoma (UPS). UPS is a subtype previously known as malignant fibrous histiocytoma. UPS is associated with one of the worst prognoses of STSs and is frequently diagnosed as a high-grade tumor. Metastatic forms of this cancer have a very low 5-year survival rate of about 16%. There is also still little understanding of the UPS subtype which involves various cellular backgrounds and mutations.
Immunotherapy continues to be a strong candidate for cancer treatment in recent years. It has already shown strong results for certain cancer types that did not see effective treatment with previously traditional treatment methods. While the immunotherapy options for STS can be limited due to tumor complexity, the UPS subtype appears to show encouraging results from immunotherapy, specifically with PD-1/PD-L1 interactions. However, there is still limited availability of patient-derived UPS cell lines which has constrained the amount of preclinical research conducted SO far.
The SCC649 JBT-19 undifferentiated pleomorphic sarcoma line is a strong option for continuing UPS research. The JBT-19 cell line appears to be the first introduced UPS cell line to simultaneously express PD-L1 and collagen, two important ligands of known immune checkpoint molecules, PD-1 and LAIR-1. This makes the cell line a strong candidate for immunotherapeutic research. The cell line can grow in both 2D and 3D formats in vitro as well as being able to produce solid tumors in mice when inoculated in vivo.
The JBT-19 cell line was established from a solid UPS tumor of a 65-year-old male. A combination of tumor fragmentation-dissociation techniques was used to establish the cells in an adherent monolayer.

Application
- Each vial contains > 1X106 viable cells.
- Cells are tested negative for infectious diseases by an Essential CLEAR Panel by Charles River Animal Diagnostic Services.
- Cells are verified to be of human origin and negative for interspecies contamination from mouse, rat, Chinese hamster, Golden Syrian hamster, and nonhuman primate (NHP) as assessed by a Contamination Clear panel by Charles River Animal Diagnostic Services
- Cells are negative for mycoplasma contamination.

Preparation Note
JBT-19 cells should be stored in liquid nitrogen. The cells can be cultured for at least 10 passages without significantly affecting cell marker expression and function.
