| Background | The adaptor protein complex 3 (AP-3) is a ubiquitously expressed, heterotetramer that is comprised of two large adaptins (AP3D1 and either AP3B1 or AP3B2), a medium adaptin (AP3M1 or AP3M2), and a small adaptin (AP3S1 or AP3S2). The AP-3 complex is involved in the sorting of protein cargo within the endosomal compartment that are destined for lysosomes (1,2). Defects in the AP-3 complex have been associated with Hermansky-Pudlak syndrome (HPS), a genetic disorder characterized by abnormal skin pigmentation, blood clotting abnormalities, and pulmonary fibrosis. Mutations in the gene encoding ubiquitously expressed AP3B1/β3A, destabilize the protein and the entire AP-3 complex, which results in defective trafficking of lysosomal membrane proteins (3,4). Research studies have shown that certain viral proteins have evolved to co-opt the AP-3 complex by binding to AP3B1 in order to facilitate viral particle assembly (5). |
