General description
Lipopolysaccharide-induced tumor necrosis factor-alpha factor (UniProt: Q99732; also known as LPS-induced TNF-alpha factor, Small integral membrane protein of lysosome/late endosome, p53-induced gene 7 protein) is encoded by the LITAF (also known as SIMPLE, PIG7) gene (Gene ID: 9516) in human. LITAF/SIMPLE is a 161-amino acid cellular protein and the C-terminus of the protein contains a highly conserved domain termed the SIMPLE-like domain (SLD). The N-terminus of SIMPLE contains two PPXY motifs that mediate protein-protein interactions with WW-domain containing proteins. The protein is known to be ubiquitously and abundantly expressed with predominant expression in the placenta, peripheral blood leukocytes, lymph nodes and spleen. Several studies have demonstrated that SIMPLE localizes to the trans-Golgi network (TGN), early endosomes, lysosomes, multi vesicular bodies, aggresomes and the plasma membrane. Three isoforms of SIMPLE have been described that are produced by alternative splicing. In a study it has been demonstrated that SIMPLE binds specifically to PI4P through its SLD domain and it co-localized with PI4P and Rab11, a marker for recycling endosomes (REs, organelles enriched in PI4P) in both the IMS32 mouse Schwann cell line and Hela cells. It is also indicated that SIMPLE may regulate protein trafficking physiologically by localizing to the TGN and/or REs by binding PI4P. LITAF/SIMPLE regulates autophagy in mature B-cell lymphomas and is also known to be a mediator of local and systemic inflammatory responses and is locally upregulated in many inflammatory diseases.
