General description
Fibromodulin (UniProt: Q06828; also known as FM, Collagen-binding 59 kDa protein Keratan sulfate proteoglycan fibromodulin (KSPG fibromodulin)) is encoded by the FMOD (also known as FM, SLRR2E) gene (Gene ID: 2331) in human. Fibromodulin is a class II small leucine-rich proteoglycan (SLRP) that plays a significant role in the extracellular matrix (ECM). Structurally, fibromodulin consists of leucine-rich repeat (LRR) motifs that are crucial for its interaction with collagen fibrils. It binds to ECM structural components and regulates collagen cross-linking, packing, assembly, and fibril architecture, impacting tissue integrity and repair, influencing the mechanical properties of tissues such as tendons, cartilage, and skin. It also plays a role in cell proliferation, differentiation, adhesion, migration, invasion, and metastasis by interacting with cytokines and growth factors, particularly with TGFβ superfamily members. Fibromodulin is predominantly expressed in connective tissues, including tendons, ligaments, and cartilage. Fibromodulin has been implicated in disease such as osteoarthritis, where altered expression or function of fibromodulin can lead to impaired collagen organization and joint degradation. Fibromodulin is a prevalent stromal component in soft tissue sarcomas and has been implicated in fibrosis and certain cancers, where it may influence tumor progression and metastasis by affecting the ECM composition and cell behavior. Preparation Note
