Claudin-4 (UniProt: O14493; also known as Clostridium perfringens enterotoxin receptor, CPE-R, CPE-receptor, Williams-Beuren syndrome chromosomal region 8 protein) is encoded by the CLDN4 (also known as CPER, CPETR1, WBSCR8) gene (Gene ID: 1364) in human. Claudin-4 is a member of the claudin family of integral membrane proteins that are essential components of tight junctions in epithelial and endothelial cells. Claudin-4 is characterized by four transmembrane domains, two extracellular loops, and cytoplasmic N- and C-termini. This protein plays a crucial role in maintaining cell polarity and regulating paracellular ion permeability. Claudin-4 is a barrier-forming claudin. It is the major claudin involved in epithelial tight junction formation in the intestines and lungs. Through interactions with other tight junction proteins and cytoskeletal elements Claudin-4 is able to influence barrier function and cell motility and regulate structure and function of other claudins. Dysregulation of claudin-4 expression is implicated in various cancers, including ovarian and breast cancer, where it is often overexpressed, correlating with increased tumor invasiveness and resistance to chemotherapy. Recent research has highlighted its involvement in pathways related to DNA repair, suggesting that claudin-4 may serve as a therapeutic target to enhance the efficacy of DNA-damaging agents. Quality Control Testing

Evaluated by Western Blotting in HT-29 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Claudin-4 in HT-29 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Claudin-4 in SW480 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound with Claudin-4 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Claudin-4 in human small intestine tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Claudin-4 in HT-29 cells.

Clone 1I21 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects Claudin-4. It targets an epitope within 19 amino acids from the C-terminal cytoplasmic domain.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
