General description
Apoptosis regulator Bcl-2 (UniProt: P10415; also known as Bcl2) is encoded by the BCL2 gene (Gene ID: 596) in human. Bcl-2 (B-cell lymphoma 2) is a pivotal anti-apoptotic protein belonging to the Bcl-2 family, which regulates cell survival and programmed cell death. Bcl-2 contains four Bcl-2 homology domains (BH1-BH4) and a transmembrane domain that anchors it to intracellular membranes, primarily the outer mitochondrial membrane. Its BH3 domain is critical for interactions with pro-apoptotic proteins such as Bax and Bak, inhibiting their activity and preventing mitochondrial outer membrane permeabilization and cytochrome c release. Bcl-2 is ubiquitously expressed across various tissues, with high levels in immune cells, epithelial cells, and neurons. Bcl-2 interacts with numerous proteins, including BH3-only proteins such as Bad and Bid, as well as non-Bcl-2 family proteins like Beclin-1, modulating autophagy and apoptosis pathways. Dysregulation of Bcl-2 expression is implicated in multiple diseases, including cancer, where its overexpression confers resistance to apoptosis and promotes tumor survival. Bcl-2 is also involved in neurodegenerative diseases like Alzheimer′s, where its imbalance contributes to neuronal death. Therapeutically, targeting Bcl-2 with BH3 mimetics has shown promise in treating hematological malignancies by restoring apoptotic sensitivity in cancer cells.
