Gene Information

General description
H-2 class II histocompatibility antigen, A beta chain (UniProt: P14483) is encoded by the H2-Ab1 (also known as H2-iabeta) gene (Gene ID: 14961) in murine species. The mouse H-2 class II histocompatibility antigen, specifically the A beta chain (H2-Ab1), is a crucial component of the adaptive immune response, primarily involved in the presentation of peptide antigens to CD4+ T cells. The H2-Ab1 gene is located on chromosome 17 and exhibits a complex expression profile, being predominantly expressed in antigen-presenting cells such as dendritic cells, macrophages, and B lymphocytes. H2-Ab1 protein is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-27), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 28-226), a transmembrane domain (aa 227-247), and a cytoplasmic domain (aa 248-265). Dysregulation or mutations in this antigen can lead to autoimmune diseases, as seen in models of diabetes and other inflammatory conditions. The AF6-120.1 antibody has been validated by Flow Cytometry and Immunohistochemistry. It cross-reacts with H-2k and H-2u haplotypes but, does not cross-react with other haplotypes (d, f, q, r, s). Application
Quality Control Testing
Evaluated by Flow Cytometry Analysis with C57BL/6 mouse splenocytes.
Flow Cytometry Analysis: 1.0 µg from a representative lot detected MHC class II I-Ab in one million C57BL/6 mouse splenocytes.
Tested Applications
Immunohistochemistry Application: A representative lot detected MHC class II I-Ab in Immunohistochemistry applications (Wang, Z., et Al. (2004). J Immunol. 172(10):5924-30).
Flow Cytometry Analysis: A representative lot detected MHC class II I-A beta chain in Flow Cytometry application (Lorber, M.I., et Al. (1982). J Immunol. 128(6):2798-803).
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
