Envelope glycoprotein GP350 (UniProt: P03200; also known as Membrane antigen (MA)) is encoded by the BLLF1 open reading frame (Gene ID: 3783713) in Epstein-Barr Virus. Epstein-Barr Virus (EBV) is a ubiquitous gamma-herpesvirus that establishes a lifelong latent infection in human B lymphocytes, with significant implications for oncogenesis and autoimmune diseases. Recent studies have elucidated the molecular mechanisms by which EBV contributes to the pathogenesis of various cancers, including nasopharyngeal carcinoma and gastric cancer. The virus encodes latent membrane protein 1 (LMP1), which mimics a constitutively active receptor, thereby activating NF-kB and JAK/STAT signaling pathways that promote cell proliferation and survival. Moreover, EBV′s role in multiple sclerosis has been reinforced by longitudinal analyses showing a high prevalence of EBV seropositivity in affected individuals. The virus′s ability to modulate host immune responses is further evidenced by its interaction with cellular proteins like TRAF1, linking viral infection to B-cell transformation. EBV′s reactivation is also associated with increased morbidity in patients with severe illnesses such as COVID-19, where it exacerbates immune dysregulation.
