Dynamin-1 (UniProt: Q05193; also known as EC:3.6.5.5, Dynamin I) is encoded by the DNM1 (also known as DNM) gene (Gene ID: 1759) in human. Dynamin-I (DNM1) is a large GTPase involved in membrane fission events, clathrin-mediated endocytosis and other vesicular trafficking processes. It is composed of a Dynamin-type g (aa 28-294), a middle domain, a pleckstrin homology (pH) domain (aa 519-625), and a proline-rich domain (aa 825-843), which together facilitate its oligomerization and interaction with phospholipids and other proteins. Dynamin-1 is highly abundant in the brain particularly at synaptic contacts. It plays a critical role in synaptic vesicle recycling. Dynamin-I localizes to the cytosol and is recruited to the plasma membrane where it assembles into helical structures around the necks of budding vesicles, using GTP hydrolysis to drive membrane constriction and fission. Dysregulation of Dynamin-I has been implicated in several neurological disorders, including epilepsy and neurodegenerative diseases, where its altered expression or function can disrupt synaptic transmission. Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Dynamin I in SH-SY5Y cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Dynamin I in NIH/3T3, and a 1:10,000 dilution from Neuro-2a cell lysate and human brain tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound with Dynamin I peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Dynamin I in human cerebral cortex tissue sections.

Clone 2N9 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects Dynamin I. It targets an epitope within 25 amino acids from the C-terminal region.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
