The cytoplasmic tyrosine/serine kinase known as SYK (Spleen Tyrosine Kinase) is essential for signal transmission, particularly in immune cells such as B cells and microglia (brain-resident macrophages). SYK interacts with the cytoplasmic domains of CD79 subunits or DAP12 when B cell or TREM2/DAP12 receptor complexes are stimulated, respectively, via binding to their distinct phosphorylated receptor motifs, known as "p-ITAMs." SYK is subsequently activated as a result of Tyr525/526 autophosphorylation of its catalytic domain. Due to SYKs subsequent phosphorylation of downstream substrates like BTK, numerous signaling cascades and physiological reactions—including cell activation, proliferation, and survival—are triggered. SYK is implicated in cancer, central nervous system disorders, and autoimmune diseases.

The AlphaLISA SureFire Mouse Total SYK Detection Kit is a sandwich immunoassay for the quantitative detection of total SYK in cellular lysates, using Alpha Technology.
