The spliced form of X-box binding protein 1 (XBP1s) is an active transcription factor that plays a vital role in the unfolded protein response (UPR). In response to endoplasmic reticulum stress and UPR, BiP relaxes its inhibitory binding to trigger IRE1. After dimerizing and auto-phosphorylating, the IRE1 receptor initiates its mRNase activity. In order to facilitate XBP-1u mRNAs translation and subsequent translocation to the nucleus, IRE1 splices it into XBP1s. Afterward, XBP-1 functions as a transcription factor. XBP1s is an important therapeutic target in studying various diseases, including cancer, diabetes, and autoimmune and inflammatory diseases.
