| Background Information | The sodium-potassium-chloride cotransporter isoform 2 is kidney-specific and is found on the apical membrane of the thick ascending limb of Henles loop and the macula densa. It accounts for most of the NaCl resorption with the stoichiometry of 1Na:1K:2Cl and is sensitive to such diuretics as furosemide and bumetanide. Some Bartter-like syndromes result from defects in this gene. SLC12A1 is kidney specific and is essential in the regulation of ionic balance and cell volume. It is a component of an electrically silent transporter system, mediating sodium and chloride reabsorption. Defects in SLC12A1 are the cause of Bartter syndrome type 1, an autosomal recessive disorder characterized by impaired salt reabsorption in the thick ascending loop of Henle with pronounced salt wasting, hypokalemic metabolic alkalosis, and varying degrees of hypercalciuria. |
