| Background | Human CD32 groupconsists of FcγRIIa, b and c proteins that share 94-99% amino acid identity intheir extracellular domains but differ substantially in their transmembrane andcytoplasmic domains. CD32B, also known as FcγRIIb, is a low affinity receptorthat recognizes immunoglobulin gammacomplexes. It is encoded by the FCGR2Bgeneand expressed on B cells and most myeloid cells. CD32B has an intrinsiccytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM) and deliversan inhibitory signal upon ligand binding. Co-ligation of CD32B with B cells antigenreceptor (BCR) results in selective dephosphorylation of CD19 tyrosine, tyrosinephosphorylation of the ITIM in the CD32B cytoplasmic domain, and recruitment ofSH2-containing inositol 5-phosphatase (SHIP). This causes a block in calciuminflux, inhibition of B cell activation, and leads to apoptosis. CD32B is apotential target for monoclonal antibody therapy for malignancies. TargetingCD32B may represent a novel approach to immunotherapy of B-cell malignancies. |
