| Background Information | Histone H2A type 1 (UniProt P0C0S8; also known as H2A.1, Histone H2A/p) is encoded by the HIST1H2AG (also known as H2AFP, HIST1H2AI, H2AFC, HIST1H2AK, H2AFD, HIST1H2AL, H2AFI, HIST1H2AM, H2AFN) gene (Gene ID 8969) in human. Histone H2A is one of four components of the core nucleosome structure. The nucleosome represents a unit of chromatin in which DNA is wrapped around a histone octamer. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones. Histones undergo a number of post-translational modifications in response to various stimuli that may induce changes in the structure of the nucleosome and hide or expose DNA sequences. Histone H2A, in particular, undergoes acetylation on lysine 5 by Tip60 enzyme, which may promote unfolding of chromatin and transcription. In addition, H2A is phosphorylated on serine 1 and serine 139 (H2A.X) residues, which mark mitosis and gene repression and DNA damage, respectively. Gene repression may also result from the sumoylation of H2A s lysine 126 residue. In addition, ubiquitination of the H2A lysine 119 residue may play a role in spermatogenesis. Monoubiquitination of Lys-120 (H2AK119Ub) by RING1, TRIM27 and RNF2/RING2 complex gives a specific tag for epigenetic transcriptional repression and participates in X chromosome inactivation of female mammals. |
