| Background Information | Single-strand selective monofunctional uracil DNA glycosylase (UniProt: Q53HV7; also known as SMUG1, hSMUG1) is encoded by the SMUG1 gene (Gene ID: 23583) in human. SMUG1 is a base excision repair enzyme that removes uracil and oxidized pyrimidines from DNA. It recognizes base lesions in the genome and initiates base excision DNA repair. It displays greater activity toward mismatches (U/G) compared to matches (U/A). SMUG1 excises uracil (U), 5-formyluracil (fU) and uracil derivatives bearing an oxidized group at C5 [5-hydroxyuracil (hoU) and 5-hydroxymethyluracil (hmU)] in ssDNA and dsDNA, but not analogous cytosine derivatives (5-hydroxycytosine and 5-formylcytosine), nor other oxidized bases. Human and rat SMUG1 exhibit about 84% sequence homology and mouse SMUG1 has 94% homology with rat SMUG1. SMUG1 was originally identified as a monofunctional uracil-DNA glycosylase selective for ssDNA, however, later it was shown to be active for U in dsDNA. SMUG1 displays the following substrate preference: ssDNA > dsDNA (G pair) = dsDNA (A pair) at low salt concentration; and dsDNA (G pair) > dsDNA (A pair) > ssDNA at high salt concentration. SMUG1 preferentially binds to a dsDNA substrate over an ssDNA substrate when both are present. SMUG1 is shown to be a pseudouridine synthase dyskerin (DKC1) partner that contributes to rRNA quality control, partly by regulating 5-hydroxymethyluridine levels. (Ref: Matsubara, M et al. (2003).
