| Background Information | Tumor necrosis factor receptor superfamily member 16 (UniProt P07174; also known as CD271, Gp80-LNGFR, Low-affinity nerve growth factor receptor, Low affinity neurotrophin receptor p75NTR, NGF receptor, p75 ICD) is encoded by the Ngfr (also known as Tnfrsf16) gene (Gene ID 24596) in rat species. p75NTR is a single pass type I membrane protein that can bind to NGF, BDNF, NT-3 and NT-4 and mediate neuronal cell survival and death. NGF and p75NTR are considered as important mediators of pain. Levels of p75NTR are shown to increase following nerve injury and its functional inhibition suppresses injury-induced neuropathic pain in rats. In non-neural cells, p75NTR expression correlates with differentiation and dedifferentiation processes and it is transient expression is observed during development of kidney, lungs, ovary, and muscle. | Application Notes | Immunocytochemistry Analysis: A representative lot immunostained 4% paraformaldehyde-fixed, 0.1% Triton X-100-permeabilized rat sympathetic neurons by fluorescent immunocytochemistry. Upregulated staining was observed in neurons infected with p75NTR ICD-expressing adenovirus (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232). Immunofluorescence Analysis: A representative lot localized p75ICD immunoreactivity in cryosections from various stages of chick embryos fixed with 4% paraformaldehyde and permeabilized by 0.1% Triton X-100 (López-Sánchez, N., et al. (2007). Physiol. Genomics. 30(2):156-171). Immunoprecipitation Analysis: A representative lot co-immunoprecipitated NRIF with p75 from rat sympathetic neurons. Co-immunoprecipitation of exogenously expressed NRIF and p75 by this antiserum was also seen using 293 transfectant. Increased CTF and ICD fragments were immunoprecipitated from PMA-treated 293 transfectants and from BDNF-treated neurons (in the presence of proteasome inhibitor). Western Blotting Analysis: A representative lot detected p75 in rat sympathetic neurons.
