| Background Information | Stimulated by retinoic acid gene 8 protein (UniProt P70278) is encoded by the Stra8 gene (Gene ID 20899) in murine species. Stra8 is an early retinoic acid (RA) responsive gene that encodes a cytoplasmic protein expressed exclusively in gonads, where it plays a pivotal role in initiating meiosis of both female and male germ cells. In female mice, Stra8 is expressed in embryonic gonocytes during a restricted time window and is indispensable for their entry into the meiotic S phase. In males, Stra8 expression is first detected in primitive spermatogonia at P5, then becomes prominent in premeiotic (preleptotene) spermatocytes from P10 onwards. Spermatogenesis, the transformation of stem cells into millions of haploid spermatozoa, is a highly regulated process that allows a constant spermatozoa production. Undifferentiated mitotic spermatogonia first develop into committed spermatogonia to produce spermatocytes, which then undergo meiosis to form haploid spermatids. Stra8 is not only required for meiosis, but also promotes spermatogonial differentiation. In the absence of Stra8, spermatogonial differentiation is impaired, giving rise to massive accumulations of undifferentiated and early differentiating spermatogonia (Type A spermatogonia) in aged testes. Stra8-deficient germ cells in postnatal male and fetal female mice arrest just before meiosis, without entering meiotic prophase. |
