| Background Information | Rho guanine nucleotide exchange factor 6 (UniProt Q15052; also known as Alpha-Pix, COOL-2, PAK-interacting exchange factor alpha, Rac/Cdc42 guanine nucleotide exchange factor 6) is encoded by the ARHGEF6 (also known as COOL2, KIAA0006, MRX46, PIXA) gene (Gene ID 9459) in human. ARHGEF6 (alpha-Pix) belongs to a family of guanine nucleotide exchange factors (GEF) that activate the Rho family of small GTPases. ARHGEF6 acts as a Cdc42 and Rac1 GEF and forms a signling module with the adaptor GIT (GPCR kinase interactor), the GTPase effector PAK (p21 activated kinase), and the GTPase activator PIX (PAK-interacting exchange factor), facilitating the activation Rac and Cdc42 GTPases at specific subcellular locations. In contrast to the ubiquitious expression pattern of ARHGEF7 (beta-Pix), ARHGEF6 is only detected in highly motile cells, including immune cells, microvascular cells, and growing fetal neurons, where it plays an essential role in mediating cell spreading, directional migration, angiogenesis, and fetal neuron dendrite outgrowth. In addition, both ARHGEF6 and ARHGEF7 are shown to interact with the RING type E3 ubiquitin ligase c-Cbl. While ARHGEF6 is a substrate for c-Cbl-mediated proteasomal degradation, ARHGEF7 binds and antagonizes against c-Cbl-mediated EGFR proteasomal degradation. Elevated ARHGEF6 accumulation is found in human A172 and rat C6 glioma cells that do not express c-Cbl, resulting in enhanced cell migration and invasion. |
