| Background Information | Neuronal acetylcholine receptor subunit alpha-4 (UniProt P43681; also known as alpha4 nAChR subunit, cholinergic receptor, nicotinic, alpha polypeptide 4, nAChR alpha4 subunit, Neuronal nicotinic acetylcholine receptor alpha-4 subunit) is encoded by the CHRNA4 (also known as NACRA4, NACHRA4) gene (Gene ID 1137) in human. The nicotinic acetylcholine (ACh) receptors (nAChRs) are activated by acetylcholine (ACh) and nicotinic agonists, such as cytisine, 1,1-dimethyl-4-phenylpiperazinium and carbachol. The nAChRs are ligand-gated pentameric cation channels. Currently, 12 neuronal and 5 muscle nAChR subunits have been identified. The nAChRs can exist as homopentamers of five identical alpha subunits (α7, α8, or α9), alpha heteropentamers composed of more than one type of alpha subunits (α7-α10), or as alpha/beta heteropentamers consisting of various combinations of alpha (α2-α6) and beta (β2-β4) subunits. The heteromeric alpha/beta types display high ligand affinity, but are insensitive to α-bungarotoxin (αBgt), while the alpha-only homomeric and heteromeric pentamers show lower agonist affinity, but high αBgt sensitivity. The α4β2 (two α4 and three β2 subunits) and α7 (five α7 subunits) nAChRs are the predominant nAChRs in the central nervous system (CNS) among mammalian species. The alpha-4 subunit is initially produced with a signal peptide (a.a. 1-28), the removal of which yields the mature 627-amino acid membrane protein with a large extracellular domain (a.a. 29-242), four transmembrane segments (a.a. 243-267, 275-293, 309-330, 601-619), and a large cytoplasmic loop (a.a. 331-600) between the third and fourth transmembrane domains. |
