| Background Information | Nuclear receptor coactivator 2 (UniProt Q15596; also known as bHLHe75, Class E basic helix-loop-helix protein 75, hTIF2, NCoA-2, Transcriptional intermediary factor 2) is encoded by the NCOA2 (also known as BHLHE75, SRC2, TIF2) gene (Gene ID 10499) in human. Nuclear receptor coactivator 2 (NCoA-2) is a 160 kDa transcriptional coactivator for steroid receptors and nuclear receptors that acts as a coactivator of the steroid binding domain (AF-2), but not the modulating N-terminal domain (AF-1). It possesses a single NR interaction domain (NID) and two autonomous activation domains, AD1 and AD2. NCoA-2 assists nuclear receptors in the upregulation of DNA expression. In cells ligand activated nuclear receptors recruit NCoA-2 to DNA where it acylates histones to make targeted regions accessible to transcriptional machinery. Along with NCOA1 it controls energy balance between white and brown adipose tissues and also acts as a critical regulator of glucose metabolism. It is also involved in the positive regulation of the transcriptional activity of the glucocorticoid receptor NR3C1 by sumoylation enhancer RWDD3. NCoA-2 is reported to positively regulate the circadian clock by acting as a transcriptional coactivator for the CLOCK-ARNTL/BMAL1 heterodimer.
