Revvity The LANCE Ultra human IL-6 Detection Kit is designed for detection and quantitation of human interleukin-6 in cell culture media using a homogeneous TR-FRET (no-wash steps, no separation steps) assay. The 500 point kit contains enough reagents to run 500 wells in 384-well format, using a 20 uL reaction volume (15 uL of sample). The 5,000 point kit contains enough reagents to run 5,000 wells in 384-well format, using a 20 uL reaction volume (15 uL of sample). One antibody of interest is labeled with a donor fluorophore (a LANCE Europium chelate) and the second molecule is labeled with an acceptor fluorophore (ULight dye). Upon excitation at 320 or 340 nm, energy can be transferred from the donor Europium chelate to the acceptor fluorophore if sufficiently close for FRET (10 nm). This results in the emission of light at 665 nm. Interleukin 6 (IL-6) is a 22 kDa pleiotropic cytokine that acts not only on the immune system, but also affects many physiological events in various organs. IL6 exerts pro- or anti-inflammatory effects, depending on the target cell analyzed and the in vivo environmental circumstances. IL-6 is a differentiation and proliferation factor for B and T cells, and acts as a migration factor on monocytic cells. It is the major activator of acute-phase protein expression in the liver, a hematopoietic factor, and acts as a survival factor on neuronal cells.
