| Background Information | Tissue inhibitor of metalloproteinases 3 (TIMP-3) is a member of the TIMP family. The TIMPs are SO named because they are slow, tight binding endogenous inhibitors of the Matrix Metalloproteinases (MMPs). The four TIMPs have different inhibition constants for the different MMPs studied. TIMPs have been shown to have activity against the ADAMs family of proteinases. TIMP-3 is an efficient "sheddase" inhibitor, inhibiting ADAM-17 (TACE) at the low nanomolar levels seen with MMP inhibition. The other ADAMs proteinases have not yet been assayed for TIMP inhibition, but TIMP-2 seems to be much less active on ADAM-17 than isTIMP-3. TIMP-3 is thought to be constitutively produced by many cell types and is inducible in others. The TIMP-3 localization differs from the other 3 TIMPs, and is thought to be primarily deposited into the extracellular matrix. |

| Entrez Gene Number |
|
| Entrez Gene Summary | This gene belongs to the TIMP gene family. The proteins encoded by this gene family are inhibitors of the matrix metalloproteinases, a group of peptidases involved in degradation of the extracellular matrix (ECM). Expression of this gene is induced in response to mitogenic stimulation and this netrin domain-containing protein is localized to the ECM. Mutations in this gene have been associated with the autosomal dominant disorder Sorsbys fundus dystrophy. [provided by RefSeq] |

| UniProt Summary | FUNCTION: Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them. May form part of a tissue- specific acute response to remodeling stimuli. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-9, MMP-13, MMP-14 and MMP-15. SIZE: 211 amino acids; 24145 Da SUBCELLULAR LOCATION: Secreted, extracellular space, extracellular matrix. DISEASE: Defects in TIMP3 are the cause of Sorsby fundus dystrophy (SFD) [MIM:136900]. SFD is a rare autosomal dominant macular disorder with an age of onset in the fourth decade. It is characterized by loss of central vision from subretinal neovascularization and atrophy of the ocular tissues. Generally, macular disciform degeneration develops in the patients eye within 6 months to 6 years. SIMILARITY: Belongs to the protease inhibitor I35 (TIMP) family. & Contains 1 NTR domain. |
| Molecular Weight | Bands may be observed for reduced protein bands at ~ 24 kDa (unglycosylated) and ~ 30 kDa (glycosylated). Additional TIMP-3 bands MAY be visible at 50 kDa (dimer), 12 kDa, and 15 kDa (breakdown products) and sample dependent. |
