| Background Information | Histone H2A deubiquitinase MYSM, often referred to as 2A-DUB is metalloprotease that specifically deubiquitinates monoubiquitinated histone H2A. 2A-DUB regulates transcription by coordinating histone acetylation and deubiquitination, and destabilizing the association of linker histone H1 with nucleosomes. The 2A-DUB mechanism may just be one of many H2A ubiquitination proteins that have the ability to regulate transcription. 2A-DUB is also essential for cell cycle progression and gene expression. |

| UniProt Summary | FUNCTION: Metalloprotease that specifically deubiquitinates monoubiquitinated histone H2A, a specific tag for epigenetic transcriptional repression, thereby acting as a coactivator. Preferentially deubiquitinates monoubiquitinated H2A in hyperacetylated nucleosomes. Deubiquitination of histone H2A leads to facilitate the phosphorylation and dissociation of histone H1 from the nucleosome. Acts as a coactivator by participating in the initiation and elongation steps of androgen receptor (AR)-induced gene activation. SUBUNIT STRUCTURE: Component of a large chromatin remodeling complex, at least composed of MYSM1, PCAF, RBM10 and KIF11/TRIP5. Binds histones. SUBCELLULAR LOCATION: Nucleus DOMAIN: Binds double-stranded DNA via the SANT domain. The SWIRM domain does not bind double-stranded DNA. POST-TRANSLATIONAL MODIFICATION: Phosphorylated upon DNA damage, probably by ATM or ATR. SEQUENCE SIMILARITIES: Belongs to the peptidase M67A family. MYSM1 subfamily. Contains 1 MPN (JAB/Mov34) domain. Contains 1 SANT domain. Contains 1 SWIRM domain. SEQUENCE CAUTION: The sequence BAB67808.1 differs from that shown. Reason: Erroneous initiation. The sequence BAG54377.1 differs from that shown. Reason: Erroneous initiation. |
| Molecular Weight | ~100 kDa observed. Uncharacterized bands at ~26, ~35, ~49 and ~66 kDa may be observed in in some lysates. Bands at ~66 and ~26 kDa may represent isoforms resulting from alternative splicing. |
