| Background Information | Human Vascular Endothelial (VE)-cadherin is a calcium-dependent adhesion molecule strictly located at cell-to-cell junctions. VE-cadherin is present in all types of endothelium (veins, arteries, capillary and large vessels). Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells. Cadherins may thus contribute to the sorting of heterogeneous cell types. VE-cadherin may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. VE-cadherin also associates with alpha-catenin forming a link to the cytoskeleton. |

| Application Notes | Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected VE-cadherin in the cell-cell junctions of HUVEC cells. Immunohistochemistry Analysis: A 1:10-1:20 dilution of a representative lot was used by an independent laboratory in paraffin-embedded tissue sections. Buffered formalin fixation is recommended with a fixation period of no longer than 12 hours. High heat antigen retrieval in citrate buffer (Cat. No. 21545) is also suggested. Antibody can also be used to label acetone-fixed cryostat sections or cells using a immunoperoxidase staining protocol (eg. IHC Select® Detection Kit, Cat. No. DAB150) Flow Cytometry Analysis: A representative lot was used in the presence of Ca2+. Note: Use PBS + 2-5mM EDTA for cell detachment. Western Blot Analysis: Non-reducing conditions may be needed, Ca2+ required in buffer (2-5 mm). |

| Entrez Gene Summary | This gene is a classical cadherin from the cadherin superfamily and is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. An alternative splice variant has been described but its full length sequence has not been determined. |

Required for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction. SUBUNIT STRUCTURE: Interacts via cadherin 5 domain with PTPRB. Interacts with TRPC4. Interacts with KRIT1. SUBCELLULAR LOCATION: Cell junction. Cell membrane; Single-pass type I membrane protein.
