| Background Information | Integrins are a family of dimeric, transmembrane proteins that mediate cell-cell and extracellular matrix adhesion. Signals transduced by integrins play a role in many biological processes, including cell growth, differentiation, migration and apoptosis. The integrin family is composed of at least 19 alpha and 8 beta subunits that form over twenty different alpha-beta non-covalently bound dimeric combinations on the cell surface. Both of the subunits contribute to the binding of the ligand. The alpha-3 subunit (CD49c) is highly concentrated in epithelial cells where it strongly adheres to Laminin-5 and Laminin-5 induced rapid adhesion can be blocked by antibodies against the alpha-3 integrin subunit. The alpha-3 subunit exists in two different splice variants, denoted as "A" and "B". The only difference that results from this differential splicing is a total change in the cytoplasmic domain, while the extracellular domain stays the same. Knock-out mice lacking this subunit show prenatal lethality and abnormalities in the kidneys. |

| Application Notes | ELISA/RIA: A 1:1,000 dilution of a previous lot was used in Enzyme-linked Immunosorbent Assay/Radioimmunoassay. Immunoprecipitation: 5 µL of a previous lot precipitated alpha3/beta1 from 5 x 106 cells (Sherry, D.M., 2001). Immunohistochemistry/immunofluorescence: 1:500 dilution of a previous lot was used, suggest use on acetone-fixed tissue. Western Blot Analysis: 1:500 dilution of a previous lot was used. Optimal working dilutions must be determined by end user. |

| Entrez Gene Summary | ITGA3 encodes the integrin alpha 3 chain. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. Alpha chain 3 undergoes post-translational cleavage in the extracellular domain to yield disulfide-linked light and heavy chains that join with beta 1 to form an integrin that interacts with many extracellular-matrix proteins. The alpha 3 beta 1 integrin is known variously as: very late (activation) antigen 3 (VLA-3), very common antigen 2 (VCA-2), extracellular matrix receptor 1 (ECMR1), and galactoprotein b3 (GAPB3). |
