| Application Notes | In preparing full-length protein samples for Western blot or immunoprecipitation, the procedures should be performed at 4°C, with the following protease inhibitor mixture: pepstatin A (1 μg/mL), leupeptin (1 μg/mL), aprotinin (1 μg/mL), Pefabloc SC (0.2 mM), benzamidine (0.1 mg/mL), and calpain inhibitors I and II (8 μg/mL each). Immunoprecipitation Immunocytochemistry. Recommend subsequent staining with a labeled second antibody. Western blot: 1.5 to 3.0 μg/mL (1:100-1:200) using ECL Dilutions should be made using a carrier protein such as BSA (1-3%) Optimal working dilutions must be determined by the end user. |

| Immunogen | Purified peptide CNB1 from 851-867 of alpha 1B subunit of rat brain voltage-gated calcium channel (Accession Q02294; Westenbroek et al. 1992) containing N-terminal lysine and tyrosine, conjugated to KLH with gluteraldehyde (Harlow & Lane 1988). |
| Concentration | Please refer to the Certificate of Analysis for the lot-specific concentration. |
| Host | Rabbit |
| Specificity | Recognizes a 1B subunit (both LMW and HMW forms) of voltage-gated calcium channel. Does not cross react with any other calcium channel antigens tested so far. SPECIES REACTIVITIES: It is expected that the antibody will also work on mouse due to sequence homology (15/17). Other species have not been tested. |

| Entrez Gene Summary | Voltage-dependent Ca(2+) channels are multisubunit complexes found in the membrane of many excitable cells that regulate calcium entry (see MIM 601011). N-type calcium channels, which control neurotransmitter release from neurons, are dihydropyridine-insensitive and omega-conotoxin-sensitive. The alpha-1 subunit forms the pore through which calcium enters the cell, and is encoded by a family of at least 5 genes.[supplied by OMIM] |

| UniProt Summary | FUNCTION: SwissProt: Q00975 # Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1B gives rise to N-type calcium currents. N-type calcium channels belong to the high-voltage activated (HVA) group and are blocked by omega-conotoxin-GVIA (omega-CTx-GVIA) and by omega-agatoxin- IIIA (omega-Aga-IIIA). They are however insensitive to dihydropyridines (DHP), and omega-agatoxin-IVA (omega-Aga-IVA). Calcium channels containing alpha-1B subunit may play a role in directed migration of immature neurons. SIZE: 2339 amino acids; 262496 Da SUBUNIT: Multisubunit complex consisting of alpha-1, alpha-2, beta and delta subunits in a 1:1:1:1 ratio. The channel activity is directed by the pore-forming and voltage-sensitive alpha-1 subunit. In many cases, this subunit is sufficient to generate voltage-sensitive calcium channel activity. The auxiliary subunits beta and alpha-2/delta linked by a disulfide bridge regulate the channel activity. Interacts with RIMS1 and RIMBP2 (By similarity). SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein. TISSUE SPECIFICITY: Isoform Alpha-1b-1 and isoform Alpha-1b-2 are expressed in the central nervous system, but not in skeletal muscle or aorta. DOMAIN: SwissProt: Q00975 Each of the four internal repeats contains five hydrophobic transmembrane segments (S1, S2, S3, S5, S6) and one positively charged transmembrane segment (S4). S4 segments probably represent the voltage-sensor and are characterized by a series of positively charged amino acids at every third position. PTM: Phosphorylated in vitro by CaM-kinase II, PKA, PKC and CGPK (By similarity). SIMILARITY: Belongs to the calcium channel alpha-1 subunit family. & Contains 1 EF-hand domain. |
