| Application Notes | Western blot: 1:1,000 using ECL. The antibody reacts with the 250 kDa IP3 Receptor 2 protein in microsomal preparations. Suggested blocking buffer is 10% normal goat serum (or same host as your secondary antibody), 1% BSA in 0.1M PBS with 0.05% Tween 20. Suggested dilution buffer is 1% normal goat serum (or same host as your secondary antibody), 1% BSA in 0.1M PBS with 0.05% Tween 20. Preferred gel percentage is 4-12% gradient gel or 5% PAGE. For microsomal methods and blotting procedure similar antibodies can be found in: Perez, et al. J Biol. Chem. 272:23961 http://www.jbc.org/cgi/reprint/272/38/23961?ijkey=35bcf508a3025fc8bf8203ea3805122eec0caf1e ) Immunocytochemistry: 1:1,000 Immunohistochemistry: 1:1,000 overnight at 2-8°C using a fluorescently labeled secondary antibody. Suggested fixative is 4% paraformaldehyde in 0.1M PBS (one hour). Suggested permeablization method is 0.05% Triton X-100 in dilution buffer. Suggested blocking buffer is 10% normal goat serum (or same host as your secondary antibody) and 1% BSA in 0.1M PBS. Suggested dilution buffer is 1% normal goat serum (or same host as your secondary antibody) and 1% BSA in 0.1M PBS. Optimal working dilutions must be determined by the end user. |

| UniProt Summary | FUNCTION: SwissProt: Q14571 # Receptor for inositol 1,4,5-trisphosphate, a second messenger that mediates the release of intracellular calcium. SIZE: 2701 amino acids; 308078 Da SUBUNIT: Homotetramer (By similarity). SUBCELLULAR LOCATION: Endoplasmic reticulum membrane; Multi-pass membrane protein. TISSUE SPECIFICITY: The short isoform is found in skeletal muscle and heart. DOMAIN: SwissProt: Q14571 The receptor contains a calcium channel in its C-terminal extremity. Its large N-terminal cytoplasmic region has the ligand- binding site in the N-terminus and modulatory sites in the middle portion immediately upstream of the channel region. PTM: Phosphorylated on tyrosine residues (By similarity). SIMILARITY: Belongs to the InsP3 receptor family. & Contains 5 MIR domains. MISCELLANEOUS: Calcium appears to inhibit ligand binding to the receptor, most probably by interacting with a distinct calcium- binding protein which then inhibits the receptor. |
