| Background Information | Cellular tumor antigen p53 is a member of the p53 family and is found in the cytoplasm, nucleus, and endoplasmic reticulum. It functions as a tumor suppressor within a variety of tumors by either stimulating apoptosis or growth arrest in deference to cell type and physiological factors. p53 serves as a trans-activator, negatively regulating cell division during cell cycle regulation. It is also thought to be involved in the cross-over for Notch signaling. Defects in p53 expression have been implicated in several diseases including; choroid plexus papilloma, lung cancer, head/neck squamous cell carcinomas, esophageal squamous cell carcinoma, Li-Fraumeni syndrome, and hereditary adrenocortival carcinoma. Mutations of the p53 protein have some characteristic features: a) Most of them are missense point mutations giving rise to an altered protein function, and b) Many -but not all- mutant p53 proteins exhibit a common mutant structure, which can be recognized by monoclonal antibodies specific for p53 in the mutant conformation. |

| Entrez Gene Summary | This gene encodes tumor protein p53, which responds to diverse cellular stresses to regulate target genes that induce cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. p53 protein is expressed at low level in normal cells and at a high level in a variety of transformed cell lines, where its believed to contribute to transformation and malignancy. p53 is a DNA-binding protein containing transcription activation, DNA-binding, and oligomerization domains. It is postulated to bind to a p53-binding site and activate expression of downstream genes that inhibit growth and/or invasion, and thus function as a tumor suppressor. Mutants of p53 that frequently occur in a number of different human cancers fail to bind the consensus DNA binding site, and hence cause the loss of tumor suppressor activity. Alterations of this gene occur not only as somatic mutations in human malignancies, but also as germline mutations in some cancer-prone families with Li-Fraumeni syndrome. Multiple p53 variants due to alternative promoters and multiple alternative splicing have been found. These variants encode distinct isoforms, which can regulate p53 transcriptional activity. Defects in TP53 are involved in esophageal squamous cell carcinoma (ESCC) [MIM:133239]. ESCC is a tumor of the esophagus. Defects in TP53 are a cause of Li-Fraumeni syndrome (LFS) [MIM:151623]. LFS is an autosomal dominant familial cancer syndrome that in its classic form is defined by the existence of a proband affected by a sarcoma before 45 years with a first degree relative affected by any tumor before 45 years and another first degree relative with any tumor before 45 years or a sarcoma at any age. Other clinical definitions for LFS have been proposed (Ref.103 and Ref.106) and called Li-Fraumeni like syndrome (LFL). In these families affected relatives develop a diverse set of malignancies at unusually early ages. Four types of cancers account for 80% of tumors occurring in TP53 germline mutation carriers: breast cancers, soft tissue and bone sarcomas, brain tumors (astrocytomas) and adrenocortical carcinomas.
