| Background Information | ULK1 or autophagy-related protein 1 homolog (ATG1) is a serine/threonine protein kinase. It is a ubiquitously expressed protein that is highly abundant in the brain, skeletal muscle, and heart. It is involved in the signaling pathway of mTOR receptors leading to the regulation of autophagy in nutrient-deprived cells. mTOR may interact with ULK1 as a complex of ULK1/FIP200/Atg13. ULK1 may play an important role in neuronal development: Elimination of ULK1s enzymatic activity by targeted disruption of its kinase domain has blocked the outgrowth of neurons in the developing mouse brain, possibly by blocking the ULK1-mediated trafficking of neurotrophins. |

| UniProt Summary | FUNCTION: Serine/threonine-protein kinase involved in autophagy in response to starvation. Acts upstream of phosphatidylinositol 3-kinase PIK3C3 to regulate the formation of autophagophores, the precursors of autophagosomes. Part of regulatory feedback loops in autophagy: acts both as a downstream effector and negative regulator of mammalian target of rapamycin complex 1 (mTORC1) via interaction with RPTOR. Activated via phosphorylation by AMPK and also acts as a regulator of AMPK by mediating phosphorylation of AMPK subunits PRKAA1, PRKAB2 and PRKAG1, leading to negatively regulate AMPK activity. May phosphorylate ATG13/KIAA0652 and RPTOR; however such data need additional evidences. Plays a role early in neuronal differentiation and is required for granule cell axon formation. CATALYTIC ACTIVITY: ATP + a protein = ADP + a phosphoprotein. SUBUNIT STRUCTURE: ATP + a protein = ADP + a phosphoprotein. SUBCELLULAR LOCATION: Cytoplasm > cytosol. Preautophagosomal structure.
