| Background Information | HuR, also known as Human antigen R and ELAV-like protein 1 is encoded for by the gene ELAVL1. HuR, is a member of the embryonic lethal abnormal vision (ELAV) protein family that has been shown to regulate gene expression by stabilizing mRNA. HuR has been shown to bind to poly-U elements and AU-rich elements (AREs) in the 3-UTR of many target mRNAs. Stabilization of mRNA by HuR requires binding and export of nuclear HuR-mRNA complexes to the cytoplasm. In human cells this process is induced by angiotensin II (AngII) via Protein Kinase C delta (PKC-δ) triggered phosphorylation of HuR at Serines 221 and 318. Phosphorylation at Serine 221 which is located within the hinge domain of HuR was found to be critical for stimulus-induced export of HuR (Doller et Al., 2010 Recent studies have shown that HuR is upregulated in certain cancers such as colon, breast and gastric cancers. HuR is also involved in embryonic stem cells (ESCs) differentiation. HuR preferentially binds mRNAs that are not methylated by N6-methyladenosine (m6A), stabilizing them, and thus promoting ESCs differentiation. HuR is also involved in the regulation of VEFG-A and NFkB expression in certain cell types. HuR activation and accumulation are also linked to many energy signaling molecules such as that of the PI3K/Akt pathway and AMPK (Doller et Al., 2010.
