| Background Information | Hematopoietic progenitor kinase 1 (HPK1), otherwise known as MEKKK 1 and MAP4K1, is a serine/threonine kinase belonging to the Ste20 (Sterile-20) family. It is predominantly expressed in hematopoietic cells and is involved in sigaling pathways requiring MAP kinase activation; HPK1 directly upstream of MEKK1. HPK1 forms signaling complexes with a number of adaptor proteins including Nck, SLP-76, and Grb2 family of proteins; these proteins may facilitate the tethering of HPK1 to membrane receptors such as the epidermal growth factor receptor. However, HPK1 may also interact with adaptor proteins to induce transcription. For example, interaction with CRKL, and Crk adaptor proteins activates the c-Jun N-terminal kinase (JNK) pathway leading to gene expression. HPK1 may mediate cell growth, cytokine signaling, and cell death. |
