| Background Information | The Bcl name is derived from B-cell chronic lymphocytic leukemia, and was given to genes involved in that disease. Members of the Bcl-2 family share homologous domains (BH) through which they are able to homo- and hetero-dimerize. Bcl-2 plays an important role in cytochrome c release from the mitochondria; it also binds Apaf1, inhibiting Apaf-1 association with caspase-9. Bcl-2 overexpression in indicated in a number of human cancers, including melanoma, breast, prostate, and lung, and it correlates with resistance to cancer treatment (presumably due to decreased apoptosis). Bcl-2 has also been implicated in schizophrenia and autoimmune disorders. Bcl-2 phosphorylated at Ser-70 plays a critial role in the translocation of Bax from the cytosol to the mitochondria, and this may regulate NO-induced neuronal apoptosis. The molecular mechanism of Bcl-2 phosphorylation and its relationship to Bax is largely unknown. |

Regulates cell death by controlling the mitochondrial membrane permeability. Appears to function in a feedback loop system with caspases. Inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). SUBUNIT STRUCTURE: Forms homodimers, and heterodimers with BAX, BAD, BAK and Bcl-X(L). Heterodimerization with BAX requires intact BH1 and BH2 motifs, and is necessary for anti-apoptotic activity By similarity. Also interacts with APAF1, RAF-1, TP53BP2, BBC3, BCL2L1, MRPL41 and BNIPL. Growth factor-stimulated phosphorylation on Ser-70 by PKC is required for the anti-apoptosis activity and occurs during the G2/M phase of the cell cycle. Translocation t(14;18)(q32;q21) with immunoglobulin gene regions. BCL2 mutations found in non-Hodgkin lymphomas carrying the chromosomal translocation could be attributed to the Ig somatic hypermutation mechanism resulting in nucleotide transitions.
