| Background Information | Histone-lysine N-methyltransferase NSD2 (UniProt: O96028; also known as EC:2.1.1.357, Multiple myeloma SET domain-containing protein, MMSET, Nuclear SET domain-containing protein 2, Protein trithorax-5, Wolf-Hirschhorn syndrome candidate, WHSC1) is encoded by the NSD2 (also known as KIAA1090, MMSET, TRX5, WHSC1) gene (Gene ID: 7468) in human. WHSC1 is a histone methyltransferase that methylates histone H3 at lysine 36 and is involved in the regulation of gene transcription during various biological processes. It is widely expressed, and its higher expression is observed in thymus and testis. It is reported to specifically dimethylate nucleosomal histone H3 at lysine 36 (H3K36me2) and can monomethylate nucleosomal histone H3 at lysine 36 (H3K36me) in vitro. At the euchromatic regions in embryonic stem cells (ESC) it is shown to trimethylate lysine 36 (H3K36me3). In ESC, it associates with developmental transcription factors such as SALL1 via PHD-type zinc fingers and represses inappropriate gene transcription mediated by histone deacetylation. WHSC1 is reported to play an essential role in adipogenesis, by regulating expression of genes involved in pre-adipocyte differentiation. A chromosomal aberration involving NSD2 is shown to cause multiple myeloma. Overexpression of WHSC1 is reported in a variety of cancer cells and its knockdown decreases proliferation in cancer cells at least in part by loss of H3K36me2 levels.
