| Background Information | Caspase-4 (CASP-4), also known as Caspase-11 (CASP-11), or Protease ICH-3, and encoded by the gene names Casp4, Casp11, Caspl, and Ich3, is a member of the peptidase C14A family and might be involved in ER-stress induced apoptosis. Caspase-4 protein has been shown to cleave Caspase-1 and might be involved in the activation cascade of caspases responsible for apoptosis execution. Caspase-4 promotes IL-1 beta processing by ICE, and therefore may also play a role in inflammatory responses. Caspase-4 has been indicated to interact with TMEM214, in the process of ER membrane localization. Additionally, Caspase-4 is a heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 20 kDa (p20) and a 10 kDa (p10) subunit. The two subunits are derived by an autocatalytic mechanism, from the precursor sequence. Recent studies have shown that the activity of Caspase-4 is increased 30-fold by endotoxins (LPS). Other studies have indicated that Caspase-4 is induced by ER stress in a DDIT3/CHOP-dependent manner. Caspase-4 is expressed mostly in lung and spleen, and less in liver, skeletal muscle, kidney and testis. |
