| Background Information | Gli3 can function as both a transcriptional activator and repressor of the sonic hedgehog (Shh) pathway. The full-length Gli3 form (GLI3FL) becomes an activator (GLI3A) after phosphorylation and nuclear translocation. The C-terminally truncated form (GLI3R), acts as a repressor. Proper balance between the Gli3 activator and the repressor specifies limb digit number and identity during development. In concert with TRPS1, Gli3 plays a role in activating chondrocyte proliferation. Gli3 is expressed in several normal adult tissues, including lung, colon, spleen, placenta, testis, and myometrium. Gli3 defects are associated with Greig cephalo-poly-syndactyly syndrome (GCPS), Pallister-Hall syndrome (PHS), polydactyly postaxial type A1 (PAPA1), polydactyly postaxial type B polydactyly (PAPB), and polydactyly preaxial type 4 (POP4). |
