Takara NucleoMag DNA Swab \1 x 96 Preps

The NucleoMag DNA Swab kit is designed for high-throughput purification of genomic DNA from various swab specimens, enabling efficient sample processing for a wide array of genetic testing applications. The kit has been validated with both cotton and synthetic swabs (e.g., flocked swabs), and employs a magnetic bead-based purification technology that can be readily automated on common liquid handling instruments (e.g., Hamilton, Tecan, and Eppendorf platforms) or magnetic separators such as the KingFisher Duo or Flex. Following reversible adsorption of nucleic acids to superparamagnetic beads, highly pure DNA is eluted under low-salt conditions and can be incorporated directly in downstream applications such as qPCR or NGS. The NucleoMag DNA Swab kit typically yields 1–3 µg of DNA per prep, and enables processing of 96 samples in ~30 min on the KingFisher Flex (excluding sample lysis).

Assessment of yield and qPCR performance of human genomic DNA isolated from different swab types using NucleoMag DNA Swab.Human genomic DNA was purified from buccal swab specimens obtained from >6 individuals using either cotton or synthetic swabs (COPAN FLOQSwabs), respectively. Lysates were prepared using NucleoSpin Forensic Filters and DNA was isolated using automated processing on a KingFisher Flex platform according to the standard protocol for the NucleoMag DNA Swab kit. qPCR performance was evaluated using the Quantifiler Human DNA Quantification assay.

Sensitive detection of bacterial DNA in human swab specimens using NucleoMag DNA Swab.DNA was isolated from mouth swabs on a KingFisher Flex platform using the standard protocol for the NucleoMag DNA Swab kit. For each sample processed, qPCR analysis targeting the bacterial 16S rRNA gene demonstrated sensitive detection of bacteria.
