Background	Cancer-related regulator of actin dynamics (CRAD, also CRACD, KIAA1211) regulates actin polymerization and epithelial integrity by sequestering actin capping proteins CAPZA and CAPZB. CRAD stabilizes the β-catenin/E-cadherin complex. Loss of CRAD allows β-catenin to activate wnt signaling (1).While CRAD was identified as a tumor suppressor downregulated in colorectal cancer, its expression is upregulated in other human cancers, including non-small cell lung cancer (NSCLC) and breast cancer (1-3). In NSCLC cells, downregulation of CRAD inhibits proliferation and increases the rate of apoptosis (2). Mutation of CRAD in small cell lung cancer (SCLC) may serve as a prognostic marker for overall survival (4).
