| Background | DnaJ homolog subfamily C member 7 (DNAJC7) is part of a family of DNAJ proteins that act as molecular chaperones to regulate protein maintenance and quality control via its regulation of HSP70 chaperones (1). DNAJ proteins contain a J-domain, an approximately 70 amino acid region containing 4 alpha helices. The J-domain is critical for stimulation of ATP hydrolysis of HSP70 (2). There are some 50 DNAJ family members that are grouped into 1 of 3 classes based on domain structures. DNAJC7 is a Class III DNAJ protein (3). The human gene that encodes the protein, DNAJC7, is genetically linked to amyotrophic lateral sclerosis (ALS), a late-onset neurodegenerative disease characterized by degeneration of motor neurons (4,5). As quality control proteins, DNAJ dysfunction may result in aberrant folding of newly synthesized polypeptides or altered clearance of degraded proteins, contributing to disease pathology. |
